| I. Background | |
| 糖皮質(zhì)激素誘導(dǎo)的腫瘤壞死因子受體相關(guān)蛋白（GITR，也稱為 TNFRSF18 或 CD357），屬于腫瘤壞死因子受體超家族（TNFRSF），它在多種免疫細(xì)胞上表達(dá)，包括 效應(yīng) T 細(xì)胞和調(diào)節(jié)性 T 細(xì)胞。GITR 在調(diào)節(jié)免疫系統(tǒng)中發(fā)揮重要作用，尤其是在 T 細(xì)胞的 活化和調(diào)節(jié)性 T 細(xì)胞（Treg）的功能中。 GITR 的配體是 GITRL，通常在抗原呈遞細(xì)胞上表達(dá)。當(dāng) GITR 與 GITRL 結(jié)合后，會 觸發(fā)下游的信號通路，包括 NF-κB、MAPK 和 PI3K/AKT 通路，從而促進(jìn) T 細(xì)胞的存活、 增殖和效應(yīng)功能。同時，GITR 的激活還可能干擾調(diào)節(jié)性 T 細(xì)胞的抑制能力，進(jìn)一步解除 免疫抑制。 | |
