| I. Background | |
| 5-羥色胺受體(5-HTR)家族是介導(dǎo)血清素(5-羥色胺)信號(hào)傳遞的關(guān)鍵受體群,分為 7 大類(5-HT1-7)共 14 個(gè)亞型,包含 G 蛋白偶聯(lián)受體(GPCR) 和配體門控離子通道兩 類結(jié)構(gòu),廣泛分布于中樞神經(jīng)系統(tǒng)、外周組織及血小板,通過(guò)多維度調(diào)控情緒、認(rèn)知、心 血管、胃腸功能及疼痛感知。 5-HTR2C 受體作為 Gq 蛋白偶聯(lián)的 5-羥色胺受體亞型,在中樞神經(jīng)系統(tǒng)(尤其下丘腦、 邊緣系統(tǒng)及脈絡(luò)叢)高表達(dá),以內(nèi)源性配體 5-羥色胺(5-HT) 激活后觸發(fā) PLC-IP3/DAG- Ca 2+信號(hào)軸,通過(guò)調(diào)節(jié)神經(jīng)元興奮性與神經(jīng)遞質(zhì)釋放(如抑制多巴胺能通路)深度參與食 欲調(diào)控、情緒穩(wěn)定性及癲癇閾值維持。5-HTR2C 功能受獨(dú)特的 RNA 編輯機(jī)制調(diào)控,臨床 研究發(fā)現(xiàn)該受體基因(HTR2C)的功能缺失性突變與暴食癥、抗抑郁藥耐藥性抑郁癥及嬰 兒癲癇綜合征(Dravet 綜合征) 強(qiáng)相關(guān),而選擇性激動(dòng)劑(如 lorcaserin) 曾用于肥胖治 療(通過(guò)激活下丘腦 POMC 神經(jīng)元增強(qiáng)飽腹感),反向激動(dòng)劑(如 SB-242084) 則作為潛 在抗抑郁靶點(diǎn)進(jìn)入Ⅱ期試驗(yàn),凸顯其作為神經(jīng)精神代謝疾病關(guān)鍵節(jié)點(diǎn)的雙重治療價(jià)值。 |
